Biotium unveils streamlined antibody cocktails for extracellular vesicle phenotyping
Biotium has launched its ExoBrite™ CD9/CD63/CD81 Antibody Cocktails, targeting three widely utilised extracellular vesicle (EV) markers to simplify flow cytometry workflows in clinical laboratories and research settings. The new reagents promise enhanced signal quality whilst reducing optimisation requirements for multi-marker EV analysis.
Dual-format approach addresses diverse analytical needs
The antibody cocktails are available in two distinct configurations: a single-colour option designed for high-coverage EV staining and a 3-colour variant optimised for phenotyping analysis. The 3-colour format offers two spectral combinations – blue/green/far-red and green/orange/far-red – providing flexibility for existing cytometry panels, whilst the single-colour option is available in green, orange, or far-red formats.
“ExoBrite tetraspanin antibody conjugates are popular tools for EV labelling for flow cytometry, and we know many of our customers combine them and stain all three tetraspanins simultaneously,” explains Dr Alexis Madrid, head of Biotium’s EV product development group. “This inspired us to create optimised tetraspanin antibody cocktails to make it even easier for our customers to stain EVs with their preferred workflow.”
Technical specifications emphasise signal integrity
The cocktails target CD9, CD63, and CD81– established tetraspanin markers for EV characterisation – using fluorophore-conjugated antibodies that maintain the performance standards of Biotium’s individual ExoBrite Flow Antibody Conjugates. A proprie-
tary formulation reduces antibody aggregation, addressing a common challenge in multi-parameter EV analysis that can compromise signal-to-noise ratios.
Unlike conventional antibodies that may lack EV-specific validation, ExoBrite Flow Antibodies undergo testing specifically on isolated EVs, ensuring reliable tetraspanin profiling. The low-aggregation formulation delivers enhanced accuracy in staining protocols, particularly relevant for clinical applications requiring consistent reproducibility.
Integration with existing analytical platforms
The antibody cocktails are designed for compatibility with multi-target panels and can be paired with ExoBrite True EV Membrane Stains or ExoBrite EV Surface Stains for expanded multi-parameter analysis. For western blot applications, Biotium offers complementary fluorescent ExoBrite Western Antibodies against CD9, CD63, and CD81 for high-sensitivity detection in EV extracts.
The cost-efficient 3-in-1 mixture approach reduces reagent consumption and preparation time whilst maintaining analytical precision – considerations particularly relevant for high-throughput clinical laboratory environments.
