Discriminating antibody panels for rare event detection

The DuraClone RE* antibody panels provide tools to study low frequency populations of either abnormal CD5+ B cells (RE CLB), abnormal plasma cells (RE PC) or abnormal B progenitor cells (RE ALB). The three new standardized, pre-formulated DuraClone RE* kits deliver high sensitivity for rare cell populations, able to accommodate either large sample volumes or high cellular concentrations. Using dry, unitized antibody combinations helps preserve the strict assay conditions essential for reliable, reproducible rare event analysis. The DuraClone family of dry reagents simplifies sample preparation and handling.  The panels eliminate errors from manual antibody pipetting, streamline workflow, improve turnaround time (TAT) and simplify inventory management. The kits are stable at room temperature, even when opened, and easy to transfer between laboratories. The first Duraclone standardized dry, pre-formulated panels were introduced in 2014, to support research into the human immune system.  Since then, the company has added DuraClone IM and IF panels for investigating immune profiles and function. While preserving strict assay conditions, which is integral to rare event analysis, the DuraClone RE* kits, produced in collaboration with experts from leading clinical research laboratories, enable streamlined workflow and standardization of results that are essential to the experimental robustness of longitudinal and multi-centric research into blood disorders. Internationally-recognized German clinical research scientists commented that the inclusion in the DuraClone RE CLB panels of ROR-1 as a new discriminative marker has markedly facilitated the screening for low frequencies of abnormal B cells and also that the DuraClone Panels provide a sensitive, analytically reliable and economic tool for the detection of minute, abnormal, plasma cell populations. DuraClone RE antibody panels are available in a convenient 25-test package containing three complete compensation kits specific for each panel. They offer the flexibility of additional antibody drop-in slots, making it possible to answer a customer’s specific questions. Further additions to the DuraClone product family are in development, including dry stimulation cocktails for functional assays.

17 OH progesterone ELISA with neonatal procedure

The new 17 OH Progesterone Elisa (# KAP1401) uses a new polyclonal antibody and provides a validated extraction procedure for newborns serum samples. Extraction of the newborns samples is highly recommended in order to avoid any potential cross reaction with the 17OH-pregnenolone sulfate present in high concentration in very young infants. The DIAsource assay has the unique advantage to provide in the CE-IVD instructions for use a validated extraction procedure in addition to the corresponding expected concentration for newborns, men and women samples. Moreover the assay offers flexibility as it can be used on serum and EDTA/Heparin plasma samples and is easily adaptable for open-system ELISA analysers. Lastly, the 24-month shelf-life allows end-users to store the kits for a long term period with no impact on the stability, quality and performance characteristics of the assay.

Fast DNA extraction for POC IVD

The Arcis DNA Blood Kit (IVD) is now available for preparation of DNA from blood using a novel three minute two step system, designed for in vitro diagnostic use. The new kit is sold in a ready-to-use format, and enables users to stabilize and extract nucleic acid from fresh or frozen whole blood samples in just three minutes, without the need for prior sample preparation steps. Arcis DNA Blood Kit is also suitable for specimens that have been stored in EDTA and heparin-containing solutions. The kit is non-hazardous, and ships and stores at room temperature for ease of handling. Designed with point-of-care in vitro diagnostics in mind, DNA extracted using the kit is also suitable for use in PCR or other applications. The new Arcis DNA Blood Kit is the first product developed specifically for use up front of IVD and point-of-care technologies. Where access to instrumentation required for traditional extraction methods is not practical, this kit’s practicality and ability to be shipped and stored at ambient temperatures has big upsides for users in emerging markets.

2-manipulation technique for SNPs detection without DNA extraction

The LAMP Human FII&FVL Duplex KIT (rs1799963/rs6025) allows every lab to perform the qualitative detection of the Prothrombin G20210A mutation (rs1799963) and the Factor V Leiden mutation (rs6025) by Loop-mediated isothermal amplification (LAMP) in an easy, fast and cost-effective way. LaCAR offers a wide panel of SNP detection kits requiring only two manipulations and without DNA extraction. The whole process including amplification, reading and results interpretation can be performed in approximatively 30 minutes. Every product developed by LaCAR has been validated and CE-IVD marked on its LC-Genie III (compact device for amplification, reading and results interpretation – see photo) but also on various high throughput platforms such as LightCycler 480 (Roche), LightCycler 96 (Roche), CFX96 (Biorad)… LAMP is a relatively new DNA amplification technique, using an isothermal nucleic acid amplification, which eliminates the need for expensive thermocyclers used in conventional PCR or qPCR and could provide major advantages due to its simplicity and ruggedness. In LAMP, the target is amplified at a constant temperature (+/- 65°C) using three sets of primers and a polymerase with high strand displacement activity. Detection of the different genotypes is performed by melting curve analysis after amplification. Both mutations are detected simultaneously by using a different fluorescent colour for each mutation.

A major software upgrade for the RAPIDComm Data Management System (V6.0) makes it possible to centrally manage in vitro diagnostics analysers and operators at the point-of-care (POC). In addition to the system’s enhanced productivity and performance features, RAPIDComm V6.0 now connects with POC cardiac devices. Point-of-care coordinators face tremendous workflow challenges when implementing and managing POC testing programmes. They are typically responsible for managing the testing done at dozens of locations on hundreds of devices and by thousands of operators, all while ensuring results are delivered quickly and reliably, and that compliance requirements of the laboratory remain satisfied. RAPIDComm V6.0 offers a complete solution for POC data management with the addition of cardiac support. With connectivity to the Stratus CS 200 Acute Care Diagnostic System, the RAPIDComm system facilitates increased accessibility and oversight for point-of-care coordinators. System features such as the lock-out component—which prohibits operators not yet certified to use the equipment from running tests—are designed to improve compliance practices and enhance patient safety. With the introduction of several performance and productivity features, RAPIDComm V6.0 also reduces manual intervention and physical transmittal of data from one device to another. RAPIDComm system V6.0 integrates with multiple host connections and includes time synchronization with the CLINITEK Status Connect System, which automatically synchronizes the instruments daily for consistency across the point-of-care environment. RAPIDComm system V6.0 also introduces a new configurable backup utility, making it easier for customers with remote database configurations to directly manage their database backup activities. Further, Material Use Reports for diabetes and urinalysis aid in inventory management, while Device Status alerts, email notifications and operator comments alert coordinators when the instruments need attention.

A new addition to the Stat Profile line of blood gas critical care analysers, Stat Profile Prime Plus, has gained CE mark certification and is now available in all CE-regulated countries. Prime Plus features maintenance-free sensor technology to provide 22 essential critical care tests including blood gases, electrolytes, metabolites, hematology, and co-oximetry. Prime Plus also features new and patented, non-lysing whole blood co-oximetry technology, along with automated quality control (QC), powerful data management, bidirectional connectivity, and extensive cybersecurity protection. Prime Plus incorporates Nova’s innovative, maintenance-free sensor technology with individual MicroSensor cards, calibrator cartridges, and quality control cartridges. This design eliminates sensor and co-oximeter maintenance, improves analyser uptime, and reduces testing costs. The MicroSensor Card provides 22 whole blood assays including co-oximetry and new critical care assays for urea, creatinine, and ionized magnesium. Prime Plus provides a complete co-oximetry panel without chemically or mechanically lysing the blood, which eliminates corrosive lysing reagents and mechanical complexity and improves instrument reliability and reduces costs. Prime Plus delivers a comprehensive critical care panel in about one minute from 135 microliters of whole blood. Tests include pH, PCO2, PO2, SO2%, Na, K, Cl, iCa, iMg, TCO2, glucose, lactate, urea, creatinine, Hct, Hb, O2Hb, COHb, MetHb, HHb, HbF, and tBil. A special safety sample port allows for easy docking and hands-free sampling, and the Clot Bloc sample flow path protects sensors from blood clot blockages and related downtime. Prime Plus provides total quality monitoring with automated QC that verifies analyser accuracy and precision on assayed liquid controls, and supplemental quality monitoring that electronically monitors and verifies correct performance of all analytical components for each analysis and calibration. It also features a powerful data management system and bidirectional connectivity, remote analyser review and control of connected Prime Plus analysers, and the most comprehensive cybersecurity features to protect patient health information and prevent viruses.

Lab-grade undercounter refrigerators use compressor-less technology

Clinical and laboratory personnel with the need to store vaccines, pharmaceuticals and other molecular or biological samples safely can benefit from a new series of lab-grade undercounter refrigerators that are designed to minimize energy usage and noise, while maintaining high levels of performance and maximizing storage capacity. As the only current lab-grade undercounter refrigerators using thermoelectric devices in place of compressor technology for variable speed control, these refrigerators regulate their internal temperatures more effectively to provide ideal storage conditions for the most demanding applications. Powered by compressor-free V-Drive technology, featuring synchronized temperature management (STeM), internal conditions are actively monitored and maintained at the desired temperature. This includes constant monitoring of, and adjustments during, door-openings to minimize the negative impact of temperature fluctuations on samples. In combination, V-Drive and STeM provide uninterrupted temperature stability and uniformity throughout the internal chamber. Furthermore, the new TSX505 refrigerators consume up to 37 percent less energy than other models, translating to an average annual cost reduction of up to 30 percent. The compressor-less technology of the TSX 505 refrigerator also overcomes the common challenge of ‘humming’ experienced with many lab-grade refrigerators through its quiet operation at just 35 dBA. This means that the unit can be kept in work areas without disturbing personnel or nearby patients.  In addition, with no internal protrusions, such as overhead fans, users can benefit from up to double the storage space compared to similar models. The undercounter refrigerators use environmentally-friendly refrigerants in compliance with the U.S. Environmental Protection Agency’s (EPA) Significant New Alternatives Policy (SNAP). Data logging capabilities are easily enabled through a USB port, allowing users to monitor storage conditions in a timely and efficient manner.
