H. pylori rapid test

Helicobacter pylori (H. pylori) is a cork-screw-spiral-like gram-negative bacterium that has adapted to grow in the human stomach. The prevalence of H. pylori infection worldwide is approximately 66%, and in developing countries, it is as high as 80% – 90%. The testing and treatment of H. pylori is very important as it has been estimated that up to 20% of people infected with H. pylori will develop ulcers which may lead to specific cancers. MP Diagnostics ASSURE H. pylori Rapid Test and Helico Blot 2.1 Western Blot incorporate a unique marker, CIM, which is used to detect the current infection of H. pylori bacteria. CIM stands for Current Infection Marker which is actually a H. pylori-specific protein. The nucleotide sequence of the CIM antigen is homologous to a conserved secreted protein. This protein has been found to be highly immunogenic for detection of H. pylori infected patients with high Positive Predictive Value of >90%. It has been coined as Current Infection Marker because IgG antibodies for CIM have been found in patients with active H. pylori infection. These patients are confirmed to have active H. pylori infection by two or more active infection diagnostic tests such as rapid urease test, culture, histology, and Urea Breath Test. With CIM-based products, no additional tests are needed for detecting current H. pylori infection.

S100A11 (S100C, Calgizzarin) is a Ca2+-binding protein of the S100 family that has been implicated in the regulation of cytoskeletal function through its Ca2+-dependent interaction with annexin I. S100A11 has been found in many different human tissues, both normal and cancerous. S100A11 appears to play distinct roles depending on the tumour involved. In bladder carcinoma or renal carcinoma, its expression is related to tumour suppression, however in prostate cancer, breast and pancreatic cancer, S100A11 is thought to be a tumour promoter. S100A11 may be a potential biomarker of infective endocarditis. Recently, increased S100A11 concentrations have also been found in patients with pulmonary diseases – COPD, idiopathic pulmonary fibrosis (IPF) and sarcoidosis. The assay has been validated to measure S100A11 in serum, bronchoalveolar lavage fluid and amniotic fluid. The 96 well-microtitre plate format ensures the applicability on standard ELISA equipment or robotic systems.

H. pylori confirmatory test

The hallmark of H. pylori infection is inflammation of the gastric mucosa. While gastritis is typically subclinical, more severe consequences of infection include mucosa associated lymphoid tissue (MALT) lymphoma, gastric carcinoma and peptic ulceration. Nearly all patients with gastric MALT lymphoma are infected with H. pylori, and the risk of developing this tumour is over six times higher in infected people than in uninfected people. Furthermore, up to eighty percent of patients with gastric MALT lymphoma achieve complete remission of their tumours after treatment with H. pylori-eradicating antibiotic therapy. Several aspects such as performance, costs, time, and feasibility should be considered in the selection of the diagnostic tests for H. pylori infection. However, invasive and intense methods are not suitable simply because of their invasive nature. The clinically accepted C-UBT test are expensive, H. pylori stool antigen tests are cumbersome and time consuming and most of the serologic tests cannot differentiate between current and non-current H. pylori infection. MP Diagnostics has developed CIM (Current Infection Marker) that detects a H. pylori specific protein. The benefits of CIM (high correlation with known gold standards, high positive predictive value of > 90% as well as detection of active H. pylori infections) provided by the ASSURE® H. pylori rapid test are also to be found in the Helico Blot 2.1 confirmatory western blot.

Besides these specialty tests, the Indiko system test menu covers routine clinical chemistry tests. The latest additions are ready-to-use liquid form CK and CK-MB reagents with a long on-board stability and wide measuring range. The unique low-volume cuvette design makes the systems economical to use, providing low reagent costs and minimizing the amount of reagent waste. The compact Indiko and Indiko Plus benchtops are designed for small and medium sized laboratory settings. Both analysers are sample-oriented and easy to use. Various automated features, like sophisticated dilutions, reflex testing capability and electronically readable calibrator and control values help to perform analysis in an effective way. Start-up and shutdown protocols are automated, and once loaded, the system has a walk-away time of up to two hours. A bi-directional LIS interface further automates test requisition and result management. The calibration and reagent lot data associated with patient information secure full result traceability.

Real-Time (quantitative) PCR assays for Chlamydia pneumonia and Mycoplasma pneumoniae  allow rapid and accurate detection of these pathogens in less than an hour run time and  with <10 copies sensitivity. There will be separate kits as well as a multiplex kit for simultaneous detection of the 2 bacteria in a single tube reaction. Detection of a genomic DNA target is included in all kits as an internal control for sample validity, extraction and amplification.  A separate positive control is included as well. These assays are adaptable to most Real-Time PCR instruments with commonly used fluorescence channels.  Bordetella pertussis & B. parapertussis RT-PCR kits are in the pipeline.
The new Bordetella pertussis IgG EIA assay, which uses pertussis toxin (PT) as antigen, as recommended in the European ECDC guidelines, and is calibrated against the international standard, complete the company’s respiratory panel, now totalling 10 different EIA kits for Mycoplasma pneumoniae, Chlamydia pneumoniae and B. pertussis.

This calibration verification is performed over the phone while the aggregometer remains in the laboratory and is available for the PAP-8E and PAP-4 Series aggregometers in good working order. This service reduces downtime, save costs, and assures the platelet aggregometer is performing to published specifications. A service technician guides the user through the calibration verification process. Required materials are shipped to the laboratory in advance of the scheduled Tele-Check appointment. Calibration results generated by the instrument are submitted by the laboratory to the company for review by a Quality Control representative. Upon approval, a Certificate of Calibration will be issued along with a Calibration Sticker.
