nSMOL for fast and precise mAb Bioanalysis

Shimadzu’s new nSMOL Antibody BA reagent kit offers a unique and standardized workflow for the quantitation of monoclonal antibody drugs by LC-MS/MS in blood or other biological samples. The technique, called nano- Surface and Molecular-Orientation Limited (nSMOL) proteolysis, has been designed to minimize sample complexity while maintaining the specificity of the protein substrate for LC–MS/MS quantitation in therapeutic drug monitoring of mAbs. nSMOL proteolysis is an entirely novel solid-solid proteolysis method engineered to deliver highly efficient and quantitative detection of CDR (Complementarity-Determining Region) peptides while decreasing the peptide numbers of the analytical target without antibody denaturation.

Uromodulin ELISA for kidney function diagnostics

A new CE-labelled Uromodulin ELISA allows measurement of the uromodulin concentration in serum or plasma, and represents a sensitive test for kidney function loss. Uromodulin is a glycoprotein which is synthesized exclusively in the kidneys and subsequently secreted. When renal function is impaired, the uromodulin concentration in the serum sinks rapidly. Thus, renal disorders can be detected at a very early stage, even when there are few symptoms. Uromodulin determination supplements established kidney function markers such as creatinine. Moreover, it enables detection of renal insufficiency in the creatinine-blind area in the initial stages of kidney disease. Uromodulin measurement is also suitable for monitoring kidney vitality during therapy, predicting clinical outcome after kidney transplantation and enhancing risk prediction for cardiovascular diseases. In the ELISA, uromodulin is detected using microplates coated with anti-uromodulin antibodies. The uromodulin concentrations are established using a simple cut-off-based interpretation (normal value in health adults around 200 ng/ml). External factors such as body weight, nutrition or muscle mass do not need to be factored into the results by additional calculations, as with classic markers. The fast and simple test procedure takes less than 3 hours and is fully automatable on all open ELISA processers.

Valveless instrumentation waste transfer pump

The new valveless Waste Transfer Pump from Fluid Metering, Inc. (FMI) was designed for removal of waste fluid generated by medical diagnostic instrumentation during the process of analysing a variety of patient fluid samples. The waste fluid generally consists of, but is not limited to, a mixture of patient samples (blood components, urine samples etc..) combined with reagents, byproducts of reagent reactions and wash fluids. These fluids can have a broad range of physical characteristics, viscosity, suspended solids, human tissue, blood cells, just to name a few. The valveless, brushless DC Waste Transfer Pump utilizes FMI’s innovative CeramPump valveless piston fluid control technology. One moving part, a rotating and reciprocating ceramic piston accomplishes all fluid control functions within the pump, thereby eliminating the need for check valves present in other reciprocating pump designs (diaphragm, bellows, solenoid piston, syringe). The variety of suspended solids and particulates present in medical instrumentation waste often causes check valves to stick, clog, and eventually fail requiring the pumps to be removed from the system and serviced. The sapphire-hard, dimensionally stable ceramic internal components of the CeramPump also substantially outperforms peristaltic pumps relying on flexible tubing. Peristaltic pumps can have integrity issues as flexible tubing over time wears and needs to be routinely replaced, at best, and at worst, can rupture resulting in bio-contaminants leaking out of the system. FMI’s valveless, brushless DC motor Waste Transfer Pump integrates the long-life performance of a brushless motor, with a built-in driver, together with the advantages of FMI’s CeramPump valveless fluid transfer technology. The pump is easily integrated into any existing system design. All that is needed is a simple 24V DC input supply. Flow rates can be factory calibrated from 20 mL/min to 400 mL/min. The integral compact design with a small 6” x 2” x 2” (approx. 15 x 5 x 5 cm) footprint makes it ideal for medical and analytical instrumentation applications.

POC test for acute respiratory infection in all-in-one format

RPS Diagnostics recently announced an updated CE mark of its FebriDx test, clearing the way for its immediate launch in the European Union and all countries recognizing the CE mark. FebriDx is a rapid, accurate, and simple to use point-of-care test that provides clinicians with a 10-minute assessment of the body’s immune response to an acute respiratory infection (ARI) directly from a fingerstick blood sample. The single use, disposable test identifies patients who have a clinically significant underlying infection and aids in the differentiation of viral and bacterial ARIs through the rapid detection of both Myxovirus resistance protein A (MxA) and C-reactive protein (CRP). MxA is an intracellular protein that becomes elevated in the presence of acute viral infection and CRP is a nonspecific inflammatory protein that is elevated in the presence of any clinically significant infection. Thus, unlike a standalone CRP test, MxA confers specificity to the test through the combined interpretation of the results.
The updated FebriDx test incorporates an all-in-one plastic housing technology that includes a built-in safety lancet, blood collection and delivery system, and integrated push button buffer delivery feature, which together, improve test convenience. The FebriDx test requires no additional equipment to perform or to interpret results. Through comprehensive analytical testing as well as a multicentre precision and reproducibility study, RPS Diagnostics successfully demonstrated equivalency to the previously CE-marked version of the product.
The timely FebriDx test results enable clinicians to formulate a targeted clinical management and therapeutic decision plan during the initial patient encounter. Antibiotic misuse is a complex global problem that leads to antibiotic resistance, avoidable adverse events, and contributes to rising healthcare costs, largely driven by diagnostic uncertainty and patient pressure. More than 50% of all antibiotic prescriptions are unnecessary and are generated in the outpatient primary and urgent care setting. In a recent study in the United Kingdom, FebriDx was shown to alter clinical management decisions in 48% of patients tested, and to reduce unnecessary antibiotic prescriptions by 80%. Each day that goes by without the implementation of a cost-effective solution is exacerbating the current global antibiotic crisis.
It is equally important not to miss treating an underlying bacterial infection as it is withholding unnecessary antibiotics from patients with clinically insignificant or viral infection. While most research and investment over the past decade has focused on rapid molecular testing to identify specific pathogens, FebriDx utilizes the body’s own immune system to provide a broad, fast, accurate, and cost-effective method to identify patients who may benefit from appropriate antibiotic therapy.

Augmented microscopy platform for pathology applications

Jennoptik Optical Systems have developed an innovative technology to bring artificial intelligence (AI) to conventional microscopy through overlay of digital content directly into a commercial-off-the-shelf microscope’s field of view, in collaboration with Google researchers. Google has demonstrated modeling for tumour detection in histopathology slides for both prostate cancer and lymph nodes from breast cancer using the platform and presented their results at the Annual meeting for the American Association for Cancer Research (AACR) in April of 2018. Jenoptik designed and integrated the opto-mechanical technology at its Jupiter, Florida location, which was made possible by leveraging their Systems Engineering experience, optical R&D capabilities, and their portfolio of IP across a breadth of innovative applications and industries. The augmented microscope solution benefitted from their optical design expertise and a digital imaging platform Jenoptik developed over the past decade.
